Effects of cholesterol, FSH and LH on steroidogenic activity of cat granulosa cells cultured in vitro

چکیده
The aim of this study was to examine the effects of 22R-hydroxycholesterol (22R-HC), folliclestimulating hormone (FSH) and luteinizing hormone (LH) on estradiol and progesterone production by cat granulosa cells. Granulosa cells from follicles were collected and cultured for up to 5 days in 24 well plates containing Dulbecco’s Modified Eagle’s Medium (DMEM)/HAM F-12 supplemented with 10 M androstenedione, 0.1% ITS premix and 0.1% bovine serum albumin, in the presence or absence of 22R-HC (10 μg/ml), FSH or LH (10, 100 ng/ml each) on first and third day. Additionally, 5% fetal calf serum was added into the culture medium for the first 24 h. Treatment of cells with 22R-HC resulted in an increase (P < 0.05) in progesterone and estradiol production on days 3 and 5 of the culture. Incubation of cells with FSH (10 and 100 ng/ml) resulted in significant stimulations of progesterone (P < 0.001) whilst incubation had no effect on estradiol production. None of the LH doses (10 and 100 ng/ml) had any effect on progesterone production by granulosa cells during the culture time. With the inclusion of 22R-HC into the culture system, progesterone synthesis was enhanced (P < 0.001) in the presence of all FSH doses.

Cellular mechanisms and modulation of activin A- and transforming growth factor beta-mediated differentiation in cultured hen granulosa cells.
Undifferentiated granulosa cells from prehierarchal (6- to 8-mm-diameter) hen follicles express very low to undetectable levels of LH receptor (LH-R) mRNA, P450 cholesterol side chain cleavage (P450scc) enzyme activity, and steroidogenic acute regulatory (StAR) protein, and produce negligible progesterone, in vitro, following an acute (3-h) challenge with either FSH or LH. It has previously bee...

The cyclical changes in the gonadotropins stimulate ovarian follicular development either to the ovulatory stage or to undergo atresia. One such intrafollicular factor may be inter-cellular communication via gap junctions. We have examined the effects of two agents (retinoids and alkanols), known to disrupt or uncouple gap junction, on FSH-stimulated LH receptor induction and progestrone synthe...
متن کاملEffects of galectin-1 on regulation of progesterone production in granulosa cells from pig ovaries in vitro.
The detection of galectin-1 (gal-1) in pig granulosa cell lysates by immunoblotting and its cytosolic as well as membrane-associated localization prompted us to study its effects on cell proliferation and regulation of progesterone synthesis. The lectin stimulated the proliferation of granulosa cells from pig ovaries cultured in serum-free medium. Gal-1 inhibited the FSH-stimulated progesterone...
متن کاملIGF1R signaling is necessary for FSH-induced activation of AKT and differentiation of human Cumulus granulosa cells.
CONTEXT FSH is routinely administered to in vitro fertilization patients to induce follicle maturation. During this process, granulosa cells differentiate and acquire specific functional characteristics that are required to coordinate ovulation and oocyte maturation. OBJECTIVE The objective of the study was to gain insight into the molecular mechanisms regulating human granulosa cell differen...
متن کاملThe Effects of Lithium Chloride on Survival and Function of in Vitro Cultured Human Granulosa Cells
Purpose: To investigate the effect of lithium chloride (LiCl) on survival and functional activity of in vitro cultured human granulosa cells (GCs). Materials and Methods: GCs were isolated from follicular fluid of women undergoing IVF/ICSI. After RBCs removal, GCs were plated in a medium containing varying concentrations of LiCl including 1, 2 and 5mM for two weeks period. A culture without Li...
